| I. Background | |
| 催乳素（PRL、Prolactin）是一種經(jīng)典的垂體激素，主要由垂體的乳酸營(yíng)養(yǎng)細(xì)胞合成并 分泌。作為多效性蛋白質(zhì)，PRL 在哺乳，繁殖，滲透調(diào)節(jié)，免疫反應(yīng)，腦功能，代謝和血 管生成的許多生理過(guò)程中既充當(dāng)循環(huán)激素。也可當(dāng)做細(xì)胞因子。催乳素可以促進(jìn)促甲狀腺 激素釋放激素（TRH）、雌激素、血管活性腸肽（VIP）的分泌，但是也可以抑制多巴胺 的分泌。 PRL 與催乳素受體（PRLR）結(jié)合，并啟動(dòng)相應(yīng)的細(xì)胞內(nèi)信號(hào)轉(zhuǎn)導(dǎo)過(guò)程以實(shí)現(xiàn)其生物學(xué) 功能。催乳素受體和生長(zhǎng)激素受體屬于 I 型細(xì)胞因子受體超家族。PRL（催乳素）結(jié)合細(xì) 胞膜上的 PRLR 后，PRLR 二聚化并激活偶聯(lián)的 JAK2 激酶，引發(fā)自身及受體磷酸化， 進(jìn)而招募并磷酸化 STATs。磷酸化的 STATs 形成二聚體，轉(zhuǎn)位至細(xì)胞核內(nèi)調(diào)控靶基因（如 Cyclin D1），促進(jìn)乳腺發(fā)育及乳汁合成。 | |
